New York State Route 76 is located entirely within Chautauqua County. It runs north-south from North Clymer at its southern end, where it splits from New York State Route 474 to New York State Route 5 along the Lake Erie shore at Ripley.
It crosses both the Southern Tier Expressway (New York State Route 17/Interstate 86) and the New York State Thruway (Interstate 90), although without a direct interchange there (access is provided via Shortman Road, the last exit on the Thruway before the Pennsylvania state line). NY 76 is the westernmost north-south highway in the state.
