這部電影交代了《星球大战II:複製人全面進攻》和《星球大战IV:新希望》之間的故事發展。它的故事發生於《星球大战II:複製人侵略》的後三年（《星球大战IV:新希望》前十九年）。在這部電影中，Anakin Skywalker被原力的黑暗面所引誘而成為黑武士，而且參議院長白卜庭宣佈他成為銀河帝國的第一任皇帝，並且解散銀河共和國。

Padmé Amidala gives birth to Luke and Leia and subsequently dies; Obi-Wan takes Luke to Tatooine to be raised by Beru and Owen Lars; Leia is taken to be raised on Alderaan by Bail Organa and his wife. C-3PO's memory banks will be wiped. However, it is not totally clear whether R2-D2's will be or not. It is said that R2 can keep a secret so he may be allowed to keep the memories of his past encounters intact.

A great purge of Jedi is also expected, including, as actor Samuel L. Jackson put it, the "satisfying" death of Mace Windu. It is these events that will almost certainly precipitate the start of the Galactic Civil War, which will be prominent in the original classic trilogy. General Grievous, who debuted in Cartoon Network microseries Star Wars: Clone Wars, plays a pivotal role in Episode III as well.

The film promises a vast opening space battle, a visit to the wartorn Wookiee planet (Kashyyyk), and a climactic lightsaber duel, said to be the longest ever filmed. It is also said to have been produced entirely without location shooting, filmed mostly in studios (in Australia and Britain) utilizing combinations of sets and blue screen shooting, with many complex locations being created in post production using a combination of Matte paintings, model work, and CGI.

There are many unresolved questions—or more pejoratively, "plot holes"—in the storyline that are hoped, by fans, to be addressed. These plot holes were formed since Episodes IV-VI were made before Episodes I-III were even written. Thus, any introductions in Episode I and II will need to be erased to explain why they were not even mentioned in the last three episodes. These problems are:

The explanation of why some Jedi get reincarnated as force ghosts while others are not.
That the droids R2-D2 and C-3PO will have their memories erased. This is implied from their not recognizing their home planet and other characters, such as their former master Darth Vader.
That there will be some sort of purge of non-humanoid species from the Empire, at least from the army and governing heads of the empire.
How 莱娅 remembers her "real mother" in episode VI if she is presumably hidden away to be raised by the Organas right after her birth and Padme is killed shortly after she is born.
The explanation of why Jar Jar Binks, the Gungan species, and the entire planet of Naboo, pivotal in the prequels, are present in Episodes I & II but not included, or even mentioned, in IV-VI.

Furthermore, in Episodes IV-VI, Obi-Wan Kenobi gave a review on Luke and Darth Vader's past. Since much of what was revealed has not been shown in Episode I or II, Episode III is expected to portray these events.

As the last theatrical Star Wars film 'Revenge of the Sith' contains many elements that make reference to the original film trilogy and George Lucas' earlier script treatments.

The title is a reprise of Lucas' first considered title 'Revenge of the Jedi' for Episode VI.
'ROTS' opens with a space battle above a planet, the first time in a Star Wars film since Episode IV.
Anakin and Obi-Wan fly their fighter craft through tight obstacles in an enemy ship, similar to Episodes IV and VI.
Anakin and Obi-Wan are sent on a daring rescue mission against an enemy fortress but all is not as it appears (Episode IV).
The planet name Utapau was Lucas' original name for Tatooine.
In the original Star Wars script treatment the climatic battle was between Wookies and Imperial forces as in ROTS. This was adapted for Episode VI as the battle on Endor between Ewoks and Imperial Stormtroopers.
Anakin/Vader's offer to Padme to join him and rule the Empire mirrors Vader's offer to Luke in Episode V.
Anakin's prophetic vision leads him on a path to the Dark Side. Luke's vision in Episode V confirms him on the path to the Light.

Palpatine/Sidious watches from a throne his current apprentice Count Dooku and his intended new apprentice Anakin duel to the death (Episode VI) while behind them can be seen a massive space fleet battle (Episode VI).
Anakin is conflicted to choose between Palpatine/Sidious and a fellow Jedi (Episode VI).
Anakin's arm is cut off by Obi-Wan. A character has an arm or a portion of an arm cut off in each of Episodes II, IV, V and VI.
